Pregnancy & Lactation

Pregnancy category D: Positive evidence of fetal risk. Contraindicated in lactation. Women of childbearing potential must use effective contraception.

Precautions & Warnings

Monitor serum calcium and vitamin D levels regularly. Dental examinations mandatory pre-treatment. Never administer IV infusion faster than 15 minutes. Evaluate thigh pain with X-ray promptly.

Use in Special Populations

Pediatric use prohibited. Geriatric patients require renal monitoring. No dose adjustment needed for hepatic impairment. Renal impairment (CrCl ≥35 ml/min) needs no modification.

Overdose Effects

Patients who have received doses higher than those recommended should be carefully monitored. Over dosage may cause clinically significant renal impairment, hypocalcemia, hypophosphatemia, and hypomagnesemia. Clinically relevant reductions in serum levels of calcium, phosphorus, and magnesium should be corrected by intravenous administration of calcium gluconate, potassium or sodium phosphate, and magnesium sulfate, respectively. Single doses of Aclasta should not exceed 5 mg and the duration of the intravenous infusion should be no less than 15 minutes.
