Primarily prescribed for dyspeptic symptoms, it alleviates bloating, epigastric pain, and acid reflux. It also treats acute nausea caused by migraines, infections, or Parkinson’s disease medications. The drug accelerates barium transit in radiological studies, making it versatile for diagnostic and therapeutic purposes.

As a dopamine antagonist, Domperidone blocks receptors in the digestive tract, improving stomach emptying without significant CNS effects. Administered 15-30 minutes before meals, adults take 10-20 mg (10-20 ml suspension) every 6-8 hours. Pediatric doses are weight-adjusted (0.2-0.8 mg/kg).

As the pituitary gland is located outside the blood-brain barrier, domperidone may induce an increase in the plasma prolactin level. In rare case this hyperprolactinaemia may give rise to neuroendocrinological phenomena such as galactorrhoea and gynaecomastia. When the blood-brain barrier is immature (as in infants) or impaired, the possible occurrence of neurological side-effects cannot be totally excluded. Rare allergic reactions, such as rash and urticaria, have also been reported.
